维持Oct-4基因表达对小鼠胚胎干细胞分化的影响 被引量:4

摘要 通过导入外源性表达的Oct-4基因,使小鼠胚胎干细胞(ES细胞)在维甲酸诱导分化后仍继续维持Oct-4基因的表达,建立了Oct-4基因维持表达的ES-O细胞株.研究发现,在缺乏干细胞分化抑制因子LIF的情况下,Oct-4基因的维持表达本身并不能维持ES-O细胞的干细胞特性;在LIF存在时,Oct-4基因的维持表达增强了ES-O细胞维持干细胞未分化状态的能力,而且部分抑制了维甲酸诱导的细胞分化,说明Oct-4基因与LIF协同作用才能维持ES细胞的未分化状态.在细胞分化过程中,ES-O细胞倾向于向神经细胞分化,提示Oct-4基因的维持表达可能与神经外胚层分化相关.
