摘要
【目的】探讨血管紧张素 (1 7) [Ang (1 7) ]对血管紧张素Ⅱ (AngⅡ )诱导培养乳鼠非心肌细胞增殖的影响。【方法】在AngⅡ诱导培养的SD乳鼠非心肌细胞中 ,应用Ang (1 7) ,通过测定非心肌细胞DNA、蛋白质合成、细胞数目等指标 ,观察非心肌细胞增殖情况。【结果】Ang (1 7)呈剂量依赖性抑制AngⅡ诱导培养的乳鼠非心肌细胞的DNA及蛋白质合成 ,与单纯AngⅡ组相比 ,Ang (1 7) 10 -9、10 -8、10 -7、10 -6mol/L分别减少 [3 H]thymidine掺入 2 1%、31%、35 %、36 % ,减少 [3 H]Leucine掺入 15 %、2 5 %、31%、32 %。Ang (1 7)还能减少AngⅡ诱导培养的乳鼠非心肌细胞数目 (AngⅡ组吸光度 :0 86± 0 10 ;AngⅡ +Ang (1 7)组 :0 78± 0 0 9,P <0 0 5 ) ,其作用能被非选择性血管紧张素Ⅱ拮抗剂 [Sar1Thr8]AngⅡ抑制。【结论】Ang (1 7)能抑制AngⅡ诱导的乳鼠非心肌细胞增殖。
Objective To explore the influence of angiotensin-(1-7) [Ang-(1-7)] on proliferation of cultured cardiac nonmyocytes of neonatal rats induced by angiotensinⅡ(AngⅡ). Methods Ang-(1-7) was used in cultured cardiac nonmyocytes of neonatal rats induced by AngⅡ, The synthesis of DNA、protein and cell numbers of nonmyocytes were employed to observe proliferation of nonmyocytes. Results Ang-(1-7) inhibited synthesis of DNA and protein of neonatal rats nonmyocytes induced by AngⅡ in a dose-dependent manner. Compared with AngⅡgroup,[3H]thymidine incorporation was decreased by 21%,31%,35%,36% and [3H]Leucine incorporation was decreased by 15%,25%,31%,32% by Ang-(1-7)10 -9,10 -8,10 -7,10 -6 mol/L respectively. Ang-(1-7) also reduced the numbers of cultured cardiac nonmyocytes of neonatal rats induced by AngⅡ[Absorbance of AngⅡgroup: 0.86±0.10, AngⅡ+Ang-(1-7)group: 0.78±0.09, P<0.05]. The effects of Ang-(1-7) could be blocked by [Sar1 Thr8] AngⅡ. Conclusion Ang-(1-7) can inhibit proliferation of cultured cardiac nonmyocytes of neonatal rats induced by AngⅡ.
