摘要
目的探讨用EBER-1作探针原位杂交检测淋巴瘤组织中EB病毒的意义.方法对16例实验性淋巴瘤组织用EBER-1作探针原位杂交检测EB病毒,同时用免疫组化法检测EB病毒表达产物及电子显微镜检测EB病毒颗粒作为对照.结果 16例淋巴瘤组织中的所有肿瘤细胞EBER-1阳性,定位于胞核,容易辨认.免疫组化检测大多数瘤细胞表达BZLF1,少数表达LMP1 和 EBNA2,电镜证实瘤细胞核内存在EB病毒颗粒.结论用EBER-1作探针原位检测EB病毒的方法敏感,特异性高,而且细胞定位清晰.
Objective:To investigate the significance of in situ hybridization detecting EB virus in experimental lymphomas by using EBER-1 probe.Methods:Using EBER-1-ISH to detect EB virus in 16 experimental lymphomas.Applying immunohistochemical assay and electron microscopy to check the EB virus gene products and EBV particles respectively.Results:In situ hybridization revealed all tumor cells of lymphomas contained EBER-1.The positive position was in nuclei and was easy to find.Immunohistochemistry showed positive staining of BZLF1 protein in almost all tumor cells while LMP1 and EBNA2 in small number of tumor cells.EBV particles were observed by electron microscopy.Conclusion:It is sensitive and highly specific to use EBER-1-ISH to detect the infection of EB virus and the cellular fixed position is clear.
