摘要 目的探讨慢性脑灌注不足皮层小胶质细胞和星形细胞的反应。方法持久性双侧颈总动脉结扎(2VO)所致慢性脑灌注不足的大鼠模型;免疫组化法观测小胶质细胞和星形细胞;光镜和电镜观察组织病理学改变。结果从1~4个月,皮层损害加重,小胶质细胞和星形细胞的活动增多。环孢素A(CsA)治疗后,皮层损害轻微,小胶质细胞和星形细胞的活动明显减弱。结论小胶质细胞和星形细胞在慢性脑灌注不足皮质损害中具有重要的作用。 Objective To explore the reaction of microglia and astrocytes in cortex after chronic cerebral hypoperfusion(CCH). Method The CCH model was established in Wistar rats with bilateral ligation of common carotid arteries. The reaction of microglia and astrocytes in cortex was observed with immunohistochemical methods. The histopathologic changes were observed with optical microscope and electron microscope. The study were investigated from 1 to 4 monthes after CCH. Result There were extensive reaction of microglia and astrocytes and histopathologic damages in the cortex after CCH. From 1 to 4 monthes, the reactivation of microglia and astrocytes increased. Meanwhile, the cortex damages has become heavier. In cyclosporin A-treated rats, the reactivation of microglia and astrocytes were significantly reduced, the cortex damages was much less intense also. Conclusion The reactivation of microglia and astrocytes may play a important rote in the pathogenesis of cortex damages after CCH. Cyclosporin A can suppressed the reactivation of microglia and astrocytes ,and reduce cortex damages after CCH.
