摘要
目的 :观察内钙拮抗剂TMB 8对过氧化氢 (H2 O2 )介导的血管内皮细胞损伤的作用。方法 :用 10 μmol/LH2 O2 损伤内皮细胞 ,观察不同浓度TMB 8对损伤内皮细胞形态学的作用 ,检测乳酸脱氢酶 (LDH)释放量、丙二醛 (MDA)生成量、超氧化物歧化酶 (SOD)和谷胱甘肽过氧化物酶 (GSH PX)活性。结果 :TMB 8能减轻内皮细胞形态学的改变 ,明显降低LDH释放 ,抑制MDA产物生成 ,保护SOD和GSH PX的活性。结论 :TMB 8对H2 O2 损伤的内皮细胞具有保护作用。
Objective:To evaluate the effects of TMB-8 on the endothelial cell injury by hydrogen peroxide. Methods:Bovine aortic endothelial cells damaged by 10 μmol/L hydrogen peroxide were divided into six groups according to the different concerntrations of TMB-8. The lactate dehydrogenase (LDH), malondia(dehyde (MDA), superoxide dismatase (SOD) and glutathione peroxidase (GSH-Px) were assayed and morphological change of endothelial cell were recorded. Results:The morphological changes of endothelial cell and the release of LDH, MDA and reduction of activities of SOD, GSH-Px, induced by H 2O 2 were inhibited by TMB-8 in the concertration-dependent manner. Conclusion:TMB-8 may protect vascular endothelial cells against hydrogen peroxide.
