摘要
目的 研究白介素 6 (IL 6 )在6 0 Coγ辐射诱导人多发性骨髓瘤细胞株XG 6、XG 7凋亡效应中的作用及其可能的分子生物学机理。方法 运用流式细胞仪技术 (FACS)、DNA片段释放法和RT PCR ,观察了IL 6对细胞株XG 6、XG 7中Bcl xL、BaxmRNA相对表达的影响及其与辐照诱导的细胞凋亡率的相关性。结果 IL 6存在时 ,XG 6、XG 7细胞的Bcl xL、BaxmRNA表达在辐照后迅速上升 ,4h达最高峰后逐渐下降 ,2 4~ 48h恢复到照射前水平 ;不加IL 6的XG 6、XG 7细胞的Bcl xL的表达在同样实验条件未检测到 ,而这 2株细胞BaxmRNA的表达在照射后不同时间均低于相应的加IL 6组 ,但尚有明显的表达。对不加IL 6的细胞组 ,辐照后不同时间的凋亡率明显高于相应的加IL 6组 (P <0 0 5 )。结论 IL 6可通过调节Bcl xL、Bax的表达及改变BcL xL Bax二者比率 ,保护辐照诱导XG 6、XG 7的细胞凋亡。
Objective To investigate interleukin 6 involvement in the response of multiple myeloma cell lines XG-6 and XG-7 to 60 Coγ-radiation. Methods Apoptosis was confirmed by flow cytometry and quantitated by the assay of DNA fragmentation release.The expression of Bcl-xL and Bax in XG-6 and XG-7 cells after irradiation and cultured with or without interleukin 6 was monitored by RT-PCR. Results XG-6 and XG-7 cells in the presence or absence of interleukin 6 underwent apoptosis at 24 h after γ-irradiation,but in the absence of interleukin 6 they had higher rates of γ-radiation-induced apoptosis than in the presence of interleukin 6(P<0 05) and had lower levels of Bcl-xL and Bax expression. Conclusion Interleukin 6 affects the sensitivity of the cell lines to γ-radiation-induced apoptosis and can rescue cells from apoptosis by up-regulating the expression of Bcl-xL and changing the Bcl-xL/Bax ratio.
