摘要
对 ABA和 PEG诱导胡萝卜 (Daucus carota L.)体细胞胚发生及其调控体细胞胚发育的效应进行了研究。在 MS液体培养基中 (含蔗糖 30 g/ L)附加 10 μm ol/ L ABA或 10 0 g/ L PEG6 0 0 0诱导体细胞胚 ,能使其提早发生 7~ 10 d,而且能使子叶胚处于调控状态。确定了 ABA和 PEG在 MS液体培养基中 (含蔗糖 10 g/ L)调控胡萝卜体细胞胚发育的下限浓度分别为 1μmol/ L ABA,2 0 0 g/ L PEG6 0 0 0 ,细胞学观察表明此种状态的胚缺乏营养。采用双向电泳分离出与 ABA和 PEG渗透调节密切相关的特异蛋白质多肽 S1 ,分子质量 2 7.9ku,等电点 6 .
ABA and PEG effects on embryogenesis and regulation of carrotsomatic embryos have been studied.When callus are suspended in MS liquid medium (containing 30 g/ L sucrose and 1 0μmol/ L ABA or 1 0 0 g/ L PEG 60 0 0 ) ,the embryos can be promoted precociously by 7- 1 0 days and the mature embryos be prevented from germination.Culturing in MS medium (containing 1 0 g/ L sucrose) ,the regulated minimum concentrations of ABA and PEG60 0 0 are1 μmol/ L and 2 0 0 g/ L respectively.The somatic embryo is short of nutration by cytology observation.Special protein S1,closely related to the osmotic regulation of ABA and PEG,is seperated(MW2 7.9ku,PI6.0 ) .From these results,we have found the bases for researching about molecular mechanism of embryogenesis in vitro and gene expression or regulation.
