讨论了现有Tm的几种定义 :(1)Tm为DNA热变性时增色效应达 5 0 %时的温度 ;(2 )Tm为DNA热变性时有 5 0 %的DNA分子发生变性时的温度 ;(3)Tm为DNA热变性时其紫外光吸收值到最大值的一半时的温度。认为定义 (1)
Several definitions of Tm are discussed in the paper;(1)Tm is the temperature at which the hyperchromic effect of the DNA molecules heated to denature has increased to 50%; (2)Tm is the temperature at which half of the heated DNA molecules have denatured; (3)Tm is the temperature at which the absorption of UV light of the DNA molecules heated to denature has increased to half of the highest absorption of UV light. Then the first one is thought to be more appropriate.
