摘要
用经硫酸铵分级沉淀,Bio—gel P-60凝胶过滤和DEAE—离子交换层析,从黑曲霉木聚糖菌株A_(34)的麦麸培养物中,分离纯化得到5个木聚糖酶组分(Ax_1、Ax_2、Ax_3、Ax_4、Ax_5).聚丙烯酰胺凝胶圆盘电泳表明均为单一带。SDS—凝胶电泳法测得分子量分别是,Ax_1为101000,Ax_2为38000,Ax_3为3000,Ax_4为24000,Ax_5为29000.圆盘等电聚胶电泳法测定5个组分的等电点分别为5.80、4.35、5.75、5.20、4.00.热处理60分钟,Ax_1、Ax_2、Ax_3、Ax_4和Ax_5的半失活温度(℃)分别为57.6、51.5、68、52.4和56.5。各组分的A_(210)^(1cm)分别为11.3,9.7,13.6,11.5和11.4.
Five xylanases, A_(x1), A_(x2), A_(x3), A_(x4), A_(x5), have been purified from Aspergillus niger A_(34) by (NH_4)_2SO_4 fractionation, gel filtration on Bio-gel P-60, and ion exchange on DEAE-Cellulose.The xylanases were shown to be homogereous by the criteria of polyacrylamide gel electrophoresis. The molecular weights were estimated to be 101000, 38000, 30000, 24000 and 29000, with pI values of 5.80, 4.35, 5.75, 5.20 and 4.00, respectively. The temperatures for the loss of half activity (t(1/2)) in 60 minutes were 57.6°C for A_(x1),51.5°C for A_(x2),68°C for A_(x3),52.4°C for A_(x4) and 56.5°C for A_(x5). The A_(280)^(1cm) of the xylanases were determined to be 11.3, 9.7, 13.6, 11.5, 11.4, respectively.
