摘要
光镜及电镜观察发现,经三七皂甙R_180μg/ml处理 5日,可诱导68%的HL-60细胞分化,其中晚幼粒32%,杆状30%,分叶6%。证实了三七皂甙R_2是HL-60细胞系的强诱导剂,可诱导HL-60细胞向中性粒细胞分化。~3H-TdR掺入实验结果表明,三七皂甙R_1在诱导HL-60细胞分化的同时,对细胞DNA和RNA的合成均有影响。与对照相比,48小时掺入抑制率~3H-TdR为26.32%,~3H-UR为18.57%(P<0.05);72小时掺入抑制率分别为17.46%和21.76%(P<0.05)。
Our experiments haveproved that human promyelocytic leuke-mia cells (HL-60) could be induced todifferentiate in vitro by notoginsenosideR_1 into mature neutrophils.The morpho-logical results showed that 68% of HL-60cells were differentiated in 8oμg/ml in-ducer for 5 days, in which metamyelo-cytes were 32%, banded neutrophils 30%and segmented neutrophils 6% ~3H-TdRand ~3H-UR incorporate assay showed thatnotoginoside R_1 effected synthesis ofDNA and RNA while it induced differen-tiation of HL-60 cell. At 48 h, percent ofincorporate inhibition was 26.32% with^3H-TdR and 18.57% with ~3H-UR; at 72 h.17.46% and 21.76%, respectively.
