摘要
用磷脂酰肌醇特异性磷脂酶C水解磷脂酰肌醇激酶催化产生的磷脂酰肌醇-4-磷酸,产物1,4-二磷酸肌醇再经碱性磷酸酶水解后,测定磷的含量,建立了磷脂酰肌醇激酶的非同位素酶解测定法。该法回收率可达91％,CV=6.5％。
A nonisotopic enzymolysisassay method of phosphatidylinositol kinase(PIK) has been developed. phosphatidy-linositol 4-phosphate(PIP), phosphorylatdfrom phosphatidylinositol(PI) by PIK washydrolyzed with phosphainositide-specificphospholipase C. The product. inositol 1,4-bisphosphate(IP_2), was separated frominositol 1-phosphate (IP) with Dowex-1column chromatography.Then, the IP_?was hydrolyzed by alkaline phosphataseto yield PI, and the PI was determined bycolorimetry. The PIK activity was definedas PI nmol/mg protein per min. Therecovery was 91%,CV=6.5%.
