用离子交换层析法纯化鸡蛋黄IgG 被引量:13

摘要 为解决鸡蛋黄IgG粗提物不纯的问题,作者试用DEAE离子交换层析法对其进行纯化。结果显示:鸡蛋黄IgG既可用0.1~0.6mol/L NaCl线性连续梯度从DEAE柱上首先洗脱出来,又可用含0.15mol/L NaCl的0.015mol/L Tris-HCl缓冲液(pH8.0)从DEAE离子交换剂上选择性地洗脱下来。采用该法制得的鸡蛋黄IgG抗体制品,具有均一、纯度高和没有明显的抗体丢失等特点,而且本法简便、可靠、实用,有益于鸡蛋黄抗体系统的开发和利用。本法在所查国内外资料中,尚未见报道。 A DEAE ion exchangechromatography procedure performed byusing either gradient or stepwise elutionwas dereloped for the purification ofchicken IgG antibodies from the egg yolkof immunized hens. Following this proce-dure,highly purified yolk IgG preparationswere obtained as identified by SephadexG 200 gel filtration as well as sodium do-decyl sulfate-polyacryamide gel electro-phoresis. Moreover,this procedure guaran-teed the recovery of antibodies of undi-minished activity.The results demonstratethat the procedure is simple,reliable. andpractical for the yolk IgG purification.Theestablishment of such a method is benefi-cial to the production and application ofegg yolk antibodies.
