摘要
本文对基因重组粒细胞集落刺激因子(rhG-CSrF)制剂用Nrs-60细胞株培养法进行了生物活性测定,结果显示rhG-CSF的对数剂量与反应呈较好的直线关系,相关系数(r)为0.991,这一结果同样也反映在另一不同基因重组技术工艺生产的rhG-CSF制剂上,由于细胞复苏传代1次与3次所得剂量反应曲线均呈很好的直线关系,因而可使本法的实验周期更短。试验结果表明NFS-60细胞株法可作为一种专属和灵敏的测定rhG-CSF活性的方法。
he bioassay for the
preparation of recombinant human granuIocyte colony stimulating factor(rhG-CSF) by NSF-60
cells methed is studied. The dose-response with rhG-CSF is linear and the relative
co-efficient(r)is 0.991. The other different mede prOduction of rhG-CSF preparation gives a
similardose-respone curve. It show that the NSF-60 cells methed has a good adaptability on
the rhG-CSF. Be-cause the dose-response of cells with one generation and three generations
is Iinear shorter of experimen-tal peried will be obtained. The results indicate that the NFS-60
microculture system may be used as aspecific and sensitive assay method for rhG-CSF.
