摘要
对徐长卿组织培养快速繁殖技术的试验研究结果表明,NAA诱导愈伤组织效果优于IAA。而IAA较适于诱导丛生芽,BA浓度有较明显的协同作用。因此,在组织培养中激素和生长素的合理配合是达到最佳培养效果的关键。试验结果表明:诱导愈伤组织的最适培养基为Ms+NA1.0g/L+NAA1.0mg/L,诱导丛生芽的最适基为Bs+BA2.0mg/L+IAA0.5mg/L,而诱导生根的最适培养基为1/2Ms+NAA1.0mg/L.通过三阶段组织培养技术的优化,可以快速繁殖出大批种苗,有效地解决徐长卿药材繁殖速度慢的问题,这一技术可望应用于生产。
series of experiments on inducing callus, developing
buds , seedlings and rcoting were done in tissuecult ure of Cynanchum paniculatum Kitag.The
results indicated that NAA was more effective in inducing callus than IAA , but IAA had better
ef-fects on inducing buds and seedling than NAA , while BA had significant cooperative rofe
both in induc-ing callus and bud. The suitable concentration of auxin and phytohormone was
the key to obtain thebest results in tissue culture. The optimized media compcosition for
inducing callus was MS+BA 1mg/L,NAA 1.mg/L. The best me-dia composition for developing
buds and seedlings was B5+BA 2 mg/L,IAA 0.5 mg/L. The satisfac-tory rooting result of vitro
plantlets was observed on the media of 1/ 2 MS+NAA 1 mg/L. This optimized three-stage
culture method was proved to be an effective way to solve the problem ofmass and
rapid-propagation in Chinese medicinal plant-Cynanchum paniculatum Kitag.
