摘要
目的了解异常蛋白负荷对鼠肾小管上皮细胞 CD5 4(ICAM- 1)表达的影响及细胞骨架蛋白的中间丝波形蛋白 (vimentin)表达的影响。方法用 Ig G(10 m g/ m L )和不同浓度的牛血清白蛋白 (BSA) (1,15 ,30 mg/ m L )作用于体外培养的鼠肾小管上皮细胞株 (NRK 5 2 E) ,采用流式细胞仪测定细胞表面 CD5 4的平均荧光强度的变化 ;对 Ig G和 BSA刺激下的爬片细胞用免疫组化的方法检测 vimentin表达的变化。结果 Ig G能引起 CD5 4表达上调 40 .1% (P<0 .0 5 ) ,BSA(15 ,30 m g/ m L )能引起 CD5 4表达上调 2 4.8%和 2 5 .0 % ,但无统计学意义 ;Ig G和 BSA(15 ,30 m g/ m L )可刺激肾小管上皮细胞表达 vim entin(P<0 .0 5 )。结论异常蛋白负荷可诱导肾小管上皮细胞粘附分子表达上调和细胞骨架的变化 ,从而可能是导致肾小管间质炎性浸润和瘢痕化的重要因素之一。
ObjectiveTo explore the expressions of CD54 (Intercellular Adhension Molecule-1) and cytoskeleton intermediate filament protein vimentin in murine renal tubular epithelial cell line with the effect of protein-overload. MethodsOne group of renal proximal tubular epithelial cell line NRK52E was treated with IgG(10 mg/mL) or different concentrations of BSA(1,15,30 mg/mL) ,another group served as control. The expression of CD54 was identified respectively by flow cytometry (FACS). Vimentin protein expression on the slides of cells treated with or without IgG or BSA was measured by immunohistological staining. ResultsThe expression of CD54 increased with the effect of IgG by 40 1% ( P <0 05),and increased with BSA(15 or 30 mg/mL) by 24 8% or 25 0% without statistical significant difference. IgG and BSA(15, 30mg/mL) could stimulate the expression of vimentin compared with control group ( P <0 05). ConclusionProtein-overload could cause the increase of adhension molecule's expression and the change of cytoskeleton in murine renal epithelial cell line, so it may be an important factor in the process of tubular interstitial infiltration and scarring.
