摘要
目的 观察一氧化氮对神经细胞的毒性作用。方法 我们利用大鼠脑神经细胞体外原代培养 NO毒性模型 ,借助倒置显微镜观察不同深度 L- Arg对神经细胞作用。结果 不同浓度 L- Arg对神经细胞起不同作用。L- Arg浓度在 40 μM对神经细胞损伤较轻 ,介于 10~ 2 0 μM或 80~ 12 80 μM之间对神经细胞损伤较大 ,L- Arg浓度>40 μM后 ,L DH(乳酸脱氢酶 )净增加率与 L- Arg浓度的增加呈线性关系 ,具有统计学意义 (P <0 .0 1) 结论 L- Arg浓度在 40 μM时部分神经细胞受损 ,此浓度为进行药物保护实验时 L- Arg的最适浓度。L- Arg浓度 >40 μM后 ,细胞死亡率于 L-
Objective To observe the neurotoxicity of nitric oxide. Method We established the neurotoxicity model of rats cerebral primary culture neuron in vitro to observe the effect of L Arg of different concentration on the neuron by using inveryted microscope. Results The action of L Arg on neuron was correlated with its concentration. L Arg of 40μM caused slight injury on neuron, but serious damage was found in the groups from 10-20μM and from 80 to 1280μM, when L Arg was above 40μM, the net increase of LDH had positive correlation with the concentration of L Arg(P<0.01).Conclusion The neuron is partially damaged when the concentration of L Arg is 40μM, which is proper in the drug-protecting trial. When the concentration is above 40μM,neuronal mortality rate has linear correlation with the concentration of L Arg.
