摘要
本文运用 PCR- SSCP银染法检测人淋巴细胞恶性肿瘤株 82 2 6、U2 66、Raji、Jurkat、Daudi p53基因状态 ,RT- PCR半定量检测 Bcl- x L的 m RNA的相对表达 ,DNA片段释放法检测细胞受照后 2 4 h的细胞 DNA链断裂量 ,探讨了 p53基因功能状态及 Bcl- x L基因表达对细胞株 82 2 6、U2 66、Raji、Jurkat、Daudi辐射敏感性的影响。结果表明 ,淋巴细胞恶性肿瘤株 82 2 6、U2 66、Raji、Jurkat、Daudi均存在 p53基因突变 ,高表达 Bcl- x
The possible mechanism of p53 gene and the expression of Bcl xL involved in response of tumor cell lines to 60 Co γ radiation has been studied. p53 gene function,the relative expression of Bcl xL and the DNA released rates of the cells irradiated by 60 Co γ rays were analysed through PCR SSCP, RT PCR and DNA released assay, repectively. The results showed that p53 gene mutations occur in malignant lymphoid cells lines of 8226, Raji, U266, Jurkat and Daudi. The levels of Bcl xL mRNA are negatively correlative with the apoptosis rates after 60 Co γ Radiation. (
