摘要 目的 :研究不同剂量碱性成纤维细胞生长因子 (b FGF)对兔成骨细胞DNA合成、碱性磷酸酶活性的影响 ,以探讨b FGF在骨折修复过程中的作用。方法 :采用酶消化分段收集法分离培养兔成骨细胞 ,倒置相差显微镜下观察细胞形态 ,免疫组化法检测细胞ALP和Ⅰ型胶原 ,酶动力学方法检测碱性磷酸酶活性 ,应用流式细胞仪进行细胞DNA含量分析。结果 :b FGF可明显促进兔成骨样细胞的DNA合成 ,但却抑制其碱性磷酸酶的活性。结论 :b FGF对骨折修复起重要作用 ,但仍需通过与其它生长因子的协同来完善。 Objective:To study the effects of different doses of b FGF on the DNA synthesis and ALP activity of the rabbit osteoblasts,and the effect of b FGF on the bone fracture restoration. Methods The osteoblasts were isolated from the rabbit ulna using the sequential enzyme digestion technique and cultivated in vitro.The osteoblasts were examined under a inversed microscope. The ALP and type Ⅰ collagen of osteoblasts were determined by immunohistochemical staining. Flow cytometer was used in the determination of DNA contents of the cells.ALP activity was detected by enzyme dynamics method. Results.It was shown that b FGF could stimulate the DNA synthesis but inhibit the ALP activity of the osteoblasts. Conclusion b FGF plays a important role in the reparation of fracture with the cooperation of other growth factors.
