肿瘤坏死因子、白细胞介素-2、α-干扰素及合用异博定对人乳腺癌细胞多药耐药性的逆转作用研究 被引量:1

摘要 目的 检测肿瘤坏死因子、白细胞介素 2、α 干扰素及合用异博定对人乳腺癌耐药细胞系的耐药性的调节作用。方法 MTT法检测药物对MCF 7/Adr细胞的抑制率变化 ,流式细胞术检测肿瘤坏死因子、白细胞介素 2、α 干扰素及合用异博定后 ,细胞内罗丹明 12 3的药物浓度变化。结果 α 干扰素可增加耐药细胞内的药物浓度 ,α 干扰素与异博定合用比单用异博定也可增加细胞内的药物浓度。各用药组均可不同程度提高阿霉素对耐药细胞的抑制率。结论 乳腺癌化疗中 ,合用α 干扰素及异博定 。 ? Objective To investigate the modulating effect of multidrug resistance in human breast cancer MCF 7/Adr cells by tumor necrosis factor,α interferon,interleukin 2 and with verapamil.Methods Chemosensitivity of cells to doxorubicin was analysed using the MTT Assay before and after the medication.P glycoprotein function of MCF 7/Adr cells treated by verapamil was examined after accumulation of the fluorescent drug,Rhodamine 123 by flowcytometry.Results α interferon could increase drug concentration in multidurg resistant cancer cells,α interferon with verapamil also could increase drug concentration compared with using verapamil only.All drugs could increase chemosensitivity to doxorubicin on different degrees,but the content of drug couldn't be increased significantly by tumor necrosis factor and interleukin 2.Conclusions α interferon could reverse multidrug resistance of human breast cancer MCF 7/Adr cells.In chemotherapy of breast cancer,We could use α interferon and verapamil to reverse multidurg resistance.Verapamil had high cardiovascular toxicity,We could reduce the side effect of verapamil by using low dose of verapamil and α interferon.This study provides a new methods to reverse multidrug resistance in clinical application.
