摘要
目的 :通过对动脉内皮细胞的培养 ,研究高浓度氧化低密度脂蛋白 (ox- L DL )和葡萄糖在糖尿病 (DM)大血管并发症发生中的作用。方法 :采用 3H- Td R标记法 ,四甲基偶氮唑盐微量酶反应比色 (MTT)法和锥虫蓝染色法。结果 :1高浓度 ox- L DL和高浓度葡萄糖抑制内皮细胞的增殖、促进内皮细胞死亡 ,并且呈浓度依赖性 ;2高浓度葡萄糖增强 ox- L DL对内皮细胞的增殖抑制及促进其死亡的作用。结论 :在高浓度葡萄糖情况下 ox- L DL致血管内皮细胞损伤的作用加强 ,提示 DM高血糖促进了脂质异常对血管内皮的损伤 ,促进 DM大血管病变的发生、发展。
Objective:The study of the effects of increased concentrations of oxidized LDL and glucose on the diabetic macrovascular complications was carried out in cultured bovine arterial endothelial cells. Methods: 3H TdR proliferation assay, MTT assay and trypan blue assay. Results:①High concentrations of oxidized LDL and glucose caused a concentration dependent inhibition of ECs replication and increase in the death of ECs. ②The elevated glucose enhanced the effects of inhibition of ECs replication and increase in the death of ECs led by high concentrations of oxidized LDL. Conclusions:The elevated glucose enhances the damaging effects of high concentrations of oxidized LDL on arterial endothelial cells, which suggests that hyperglycemia increases the vascular endothelial damage caused by dyslipdemia in diabetes and accelerates the development of diabetic macrovascular complications.
