鸡腺胃型传染性支气管炎病毒金坛分离株核蛋白基因的克隆和测序

Cloning and Sequencing of the Nucleocapsid Gene of JT Strain of Avian Infectious Bronchitis Virus

摘要 应用逆转录多聚酶链反应技术,参照 Boursnell等发表的鸡传染性支气管炎病毒核蛋白基因序列,自行设计并合成1对引物,应用该引物,从感染IBV金坛株的第五代SPF鸡胚尿囊液中扩增出片段大小为828hp的JT株 N基因,与设计相符,表明扩增片段为 IBV JT株 N基因。将扩增片段与 pGEM- T Easy载体连接,经核酸序列测定,与Genebank中报道的IBV其它株N基因同源性较高。 One pair of primers for amplifying the nucleocapsid gene of avian infectious bronchitis virus(IBV) had been designed. The amplified fragment was 828 hp from JT strain by RT - PCR. The N gene of JT strain was inserted into pGEM - T Easy vector and identified by sequencing. The homologies of nucleie acids between, JT strain and other strains is very high.
