摘要
目的 检测苯并 (a)芘的代谢产物反式—BPDE诱发的人支气管上皮细胞系P5 3基因突变 ,探讨苯并 (a)芘在人肺癌发生中的作用。方法 用银染PCR—SSCP方法检测P5 3抑癌基因第 5、6、7、8外显子的点突变情况。结果 经反式—BPDE处理的人支气管上皮细胞系 ,2 0d后 ,P5 3基因第 8外显子出现点突变。结论 结果提示 ,P5 3基因点突变可能是苯并 (a)
Objective To explore the effects of benzo(a)pyrene[B(a)P]on the development in human lung cancer, the mutations of the P53 tumor suppressor gene were detected in human bronchial epithelial cell line (16HBE) treated with trans BPDE, a metabolite of B(a)P. Methods The point mutations in exon 5 8 of P53 tumor suppressor gene in 16HBEs were detected by PCR SSCP assay. Results Following 20 day culture, 16HBEs treated with trans BPDE showed point mutations in exon 8 of P53 tumor suppressor gene. Conclusion The results suggested that P53 gene point mutation would be an early molecuar event in lung cancer induced by B(a)P.
