水稻白叶枯病原细菌单克隆抗体杂交瘤细胞株的建立及初步应用 被引量:2

摘要 经水稻白叶病原细菌(Xanthomonas campestris pv.oryzae)菌株K-s-6-6免疫的BALB/C小鼠的脾细胞与骨髓瘤细胞SP2/0融合,经筛选和克隆化获得3株稳定分泌抗该病原细菌单克隆抗体的杂交瘤细胞株K1,K2和K31。分泌抗体稳定性试验、封闭试验、染色体数计数、交叉后应试验等表明具有株特异性;抗体分属IgG3和IgG26亚类;腹水抗体滴度在1:10^4左右,初步用于检测白叶枯病菌菌株株,病叶和病种证明特异性高,匀质均一,在理论和生产上都有实用价值。 Hybriaoma cell lines (K1,K2 ane K31) secreting monoclonal antibodieswere prepared by fusion of mouse myeloma cells (SP2/0-Agl4) and spleencells derived from BALB/C mice immunized with the preparation of Xantho-monas campestris pv.oryzae Ks 6—6 (serotype 1). The specificities of the three monoclonal antibodies were tested by ELISAmethod.The results indicated that the monoclonal antibodies secreted by K1,K2 and K31 are all specific to Ks 6—6,but not to Os 213 (serotype 2). Antibody titers of ascitic fluids were about 1:10000 when measured byELISA method.The reaction of the monoclonal antibodies to Ks 6—6 could beblocked by the presence of polyclonal rebbit antiserum.These Ks 6—8 speci-fic monoclonal antibodies did not react with other plant pathogenetic bacteriawhich belong to five genera,e.g.Xanthomonas campestris,X.campestris pv.orysicola,Erwinia carotovara,Pseudomonas solanacearum,C.sepedonicumand Agrobacterium tumefaciens.The primary experiments were,also providedthat these monoclonal antibodies are especially valuable for diagnosis of thebacterium of serotype 1,most tested bacterium collected in major rice plantingarea (Jiangsu,Zhejiang,Guangxi,Yunnan and Fujian provinces) are thatof serotype 1,the ratio of positive reaction to tested samples is 44/74,Theother advantage and its application are discussed.
