摘要
建立了维 A酸血药浓度的 H PL C测定法 ,对其软胶囊和硬胶囊在家兔体内的药动学特性进行了比较。结果表明 ,软胶囊比硬胶囊体内吸收好 ,血药峰浓度高 ,其药动学参数 Tmax分别为 2 .2 49± 0 .45 7h、2 .46 1± 0 .494h(P>0 .0 5 ) ;Cmax分别为 176 .73± 145 .79ng/ml、90 .0 3± 6 0 .93ng/m l(P<0 .0 5 ) ;AUC0 -∞ 分别为 72 4.5 2± 431.5 9ng· h· m l- 1 、46 0 .5 7± 2 75 .2 2 ng· h· ml- 1 (P<0 .0 1)
Tretinoin soft gelatin capsules and hard capsules were given orally to 10 rabbits at a dosage of 10 mg in a randomized, cross over way. The blood concentration was measured by HPLC. The results showed that the soft gelatin capsules had higher peak blood concentration than the hard ones. T max were 2.249±0.457, 2.461±0.494 h( P >0.05); C max were 176.73±145.79, 90.03±60.93 ng/ml ( P < 0.05); AUC 0 ∞ were 724.52±431.59, 460.57±275.22 ng·h·ml -1 ( P <0.01), respectively.
