β-TCP/rhBMP-2复合物的制备及其诱导成骨活性观察 被引量:4

摘要 为了检测 β-磷酸三钙 /人重组骨形态发生蛋白 - 2 (β- TCP/ rh BMP- 2 )复合物的骨诱导能力 ,将自制 β- TCP/rh BMP- 2复合物植入 45只小鼠一侧股后肌袋作为实验组 ,单纯 β- TCP植入对侧作为对照组。术后 2 4小时、 72小时、1周、 2周、 4周和 8周分别取材作组织学检查 ;1、 2、 4周取材的部分标本作碱性磷酸酶 (AL P)活性测定。结果发现实验组有大量新生软骨和骨形成 ,随着时间推移 ,生成量逐渐增多 ;对照组无软骨和骨形成 ;此外 ,实验组 AL P活性明显高于对照组 (P<0 .0 0 1) ,并且随时间推移而升高。表明β- TCP/ rh BMP- 2具有良好的骨诱导作用。 To determine the osteoinductive potential of β-TCP/rhBMP-2 composite,the β-TCP/rhBMP-2 composite was obtained by combining Beta-tricalcium phosphate (β-TCP) prepared with recombinant human morphogenetic protein-2 (rhBMP-2)and was implanted into muscle pouches in the thigh of mice β-TCPalone was implanted on the opposite site as controlsAt intervals of 24 and 72 hours and 1,2,4 and 8 weeks after the implantation,the specimens were obtained,and histologic study and alkaline phosphatase assay(1,2,4 weeks)were performedIt was found that there was a large amount of new cartilage and bone formation within the composite,and incereasing with timeMeanwhile,there was no new bone formation where β-TCP alone was implantedThe levels of alkaline phosphatase in the β-TCP/rhBMP-2 implants also were increasing with time and were higher than those in controls (P<0001)The results suggest that β-TCP/rhBMP-2 possesses heterotopic osteoinductive potential
