摘要
目的 探讨三氧化二砷 (As2 O3)对人肺巨细胞癌细胞系PLA 80 1D株细胞运动能力和尿激酶 (uPA)、尿激酶受体 (uPAR)mRNA表达的影响。方法 采用 0 5 μmol/L、1 0μmol/L及 2 0 μmol/L三种浓度的As2 O3 处理人肺巨细胞癌细胞 (PLA 80 1D株 ) ,12 0h后收取细胞 ,分别采取Boyden小室测定细胞穿膜运动能力及RT PCR方法检测细胞uPA及uPARmRNA转录表达水平。结果 经As2 O3 处理的PLA 80 1D株细胞 ,穿膜运动能力和uPA及uPARmRNA转录表达水平均显著下降 ,As2 O3 作用呈剂量依赖方式。结论 As2 O3可抑制肿瘤细胞的运动能力 ;可下调uPA、uPAR表达 。
Objective To investigate the effects of arsenic trioxide (As 2O 3) on motility and expression of uPA、 uPAR mRNA in human pulmonary giant cell carcinoma cell line PLA 801D. Methods PLA 801D cell was treated with 0 5 μmol/L、 1 0 μmol/L and 2 0 μmol/L As 2O 3 for 5 days respectively. The migrative ability of PLA 801D cell treated with As 2O 3 were detected by using the Boyden chamber assay, the expression level of uPA and uPAR mRNA by semi quantitative RT PCR. Results The migrative ability of passing through the polycarbonate filter of Boyden chamber decreased in PLA 801D treated by As 2O 3, and the inhibitory effect became more obvious with increasing concentration As 2O 3. Further investigation by RT PCR denoted that the expression level of uPA and uPAR mRNA were lower in compare with untreated cells. Conclusion The migrative ability of PLA 801D cell may be inhibited and the uPA and uPAR mRNA expression, may be lowered by As 2O 3.
