摘要
采用超声波粉碎、NP 40提取与两种互补式免疫亲和层析柱相结合的新提纯方案。结果获得了高纯度精子膜抗原 ,对经两种市售试剂盒检测的阳性标本 36份、阴性标本 2 4份进行检测 ,结果一致 ,正常人血清标本A值≤ 0 0 8。初步临床应用显示不孕症患者中 ,血清阳性率 2 3 5 2 % ,精浆阳性率 7 5 7% ,与郑州华美公司试剂盒的阳性符合率达 10 0 %。
Highly purified sperm membrane antigen was obtained by ultrasonic disintegration,extraction with NP 40 and two kinds of affinity chromatographies As detected by EIA kit,the results of 36 positive and 24 negative samples were correspondent with those of the commercial kits and the serum A values were less than 0 08 Results of preliminary trials in infertility patients showed that the positive rates in sera and sperm plasm were 23 52% and 7 57% respectively which approximated to those of Zhengzhou Huamei Co up to 100%
