去甲二氢愈创木酸对活体血管生成作用的实验研究 被引量:2

摘要 目的 观察去甲二氢愈创木酸 (NDGA)对在体血管生成的影响。方法 采用鸡胚绒毛尿囊膜 (CAM)试验 ,检测NDGA对血管生成的影响。结果  2 .5× 1 0 -6mmol以上量的NDGA均对血管生成有抑制作用 ,在实验剂量 ( 0 .5~ 5 0 )× 1 0 -6mmol范围内 ,随给药量的增加 ,其抑制作用愈加明显。采用Bouin’s液原位固定后取膜、DAB染色、普通显微镜下观察或用图像分析方法可使结果更具可信性与可比性。结论 NDGA可抑制血管生成 。 Objective To investigate the effect of nordihydroguaiaretic acid (NDGA) on angiogenesis. Methods The chicken chorioallantoic membrane (CAM) assary was used to detect the effect of NDGA on angiogenesis. Results NDGA inhibited blood vessel formation in chicken chorioallantoic membrane, with the lowest effective dose of 2.5×10 -6 mmol. This inhibitory effect was proportional to the dose of NDGA. The modified CAM assay, such as fixing CAMs in situ with Bouin's solution, staining samples with DAB, and being quantified by image analysis, could make the results more convincible. Conclusion Our study find primarily that NDGA, an inhibitor of tumor cell growth, can effectively inhibit the angiogenesis of CAM, which implies its application in the treatment of tumor or angiogenesis in other pathological conditions.
