摘要
以鸡下丘脑为实验材料，以λｇｔ１０为载体，构建了鸡下丘脑ｃＤＮＡ文库。结果表明，文库的滴度为３．８×１０６，重组率为８０％，插入片段大小集中在１．０～３．０ｋｂ之间。从ｃＤＮＡ文库中随机挑取１０个克隆，并对其ＥＳＴｓ进行了测定和初步分析。经ＢＬＡＳＴｎ和ＦＡＳＴＡ３分析后发现，４个ＥＳＴｓ在鸡中已有同源序列，４个在人或其他物种中也可以找到同源序列，有两个为未知功能基因。所测１０个ＥＳＴｓ序列均已录入ＧｅｎＢａｎｋ。
Using hypothalamus as material and gt10 as vector, chicken hypothala mus cDNA library has been constructed. The results showed that the titration o f the cDNA library we constructed was 3.8×106, and the fragment length of ins erted DNA ranged mainly from 1.0～3.0kb. The ESTs was sequenced and primarily analyzed for ten clones randomly selected from above library. After analysis w ith BLASTn and FASTA3.1, it was found that four ESTs developed matched homolog ous sequence in chicken, four also had similar sequences in human and other sp ecies, have other two sequences were function-unknown gene. The sequences of t en ESTs we studied have been loaded into GenBank.
