摘要
我们报告一种简单、快速提取和纯化质粒的方法。经过细菌培养、质粒扩增制备出清亮裂解液,经酚处理后,用Sephacryl S—1000凝胶过滤层析除去了菌体RNA。制备出的高纯度质粒DNA可用于限制内切酶等分析与应用。
We report a simple, rapid and reliable procedure for large-scale purification of plasmid DNA. After bacterial culturation and amplification of plasmids, the crude extraction of plas-mids was purified through gel-filtration chromatography using Sephacryl S-1000. The recovered plasmids are free from RNA. Such high purified plasmid DNA is suitable for restriction analysis.
