摘要
目的:初步探讨甲状腺乳头状癌病人肿瘤组织和甲状腺正常组织间,及淋巴结转移组和未转移组间的基因差异表达。方法:收集12例甲状腺乳头状癌病人的肿瘤组织及对侧甲状腺正常组织,根据肿瘤和正常组织,淋巴结是否转移进行分组,抽提总RNA,质检合格后利用基因芯片技术初步分析各组间的差异基因表达。结果:通过DiffGene算法筛选出肿瘤组和对照组,淋巴结转移组和未转移组间表达有显著差异的基因。肿瘤组和对照组间显著差异表达基因2 028条,上调1 022条,下调1 006条。结论:差异基因的筛选为进一步研究甲状腺乳头状癌发生发展的分子机制提供了研究方向和实验基础。
Objective To study the differentially expressed genes of the patients from the tissues between papillary thyroid carcinomas and normal thyroid, and between lymph node metastasis and non-lymph node metastasis. Methods There were 12 patients with papillary thyroid carcinomas in this study including 4 groups: tumor group with carcinoma, control group from contralateral thyroid, the group of patients with lymphatic metastasis, and the group with non-lymphatic metastasis. Total RNA was extracted and gene chip was used to analysis the differential gene expression between groups. Results There were significantly differentially expressed genes between tumor group and control group, as well as the group of lymphatic metastasis and that of non-lymphatic metastasis. Among tumor group and control group, there were 2 028 differentially expressed genes with 1 022 up-regulated and 1 006 down-regulated. Conclusions The screening of differentially expressed genes might be experimental basis providing the direction for further research of molecular mecha- nism of papillary thyroid carcinomas.
