摘要
目的 克隆胶质瘤相关基因 .方法 取胶质瘤患者新鲜肿瘤标本及肿瘤附近正常脑组织提取 m RNA,反转录成c DNA.采用基于 PCR的消减杂交法获得差异的 c DNA,并经多聚酶链反应扩增后克隆入 T载体测序 .用打点杂交法验证差异克隆 .结果 获得一胶质瘤相关的酸性成纤维生长因子基因同源物 .结论 该酸性成纤维生长因子基因同源物是酸性成纤维生长因子 m RNA的不同拼接产物 ,可能参与胶质瘤的形成 .
AIM To search for glioma related genes. METHODS Glioma sample and normal brain tissue of a patient were got under operation. mRNAs were extracted and reverse transcribed into cDNA. Subtractive hybridization was done to get the glioma specific cDNA which was amplified by polymerase chain reaction (PCR). The PCR products were cloned into vectors for sequencing. Dot hybridization was used to testify the different clone. RESULTS A homolog of acid fibroblast growth factor (aFGF)mRNA was found in tumor tissue but not in normal brain tissue. CONCLUSION The new gene is the different splicing product of aFGF and might be involved in glioma genesis.
