摘要
目的:研究长期高脂膳食对雄性SD大鼠精子凋亡的影响,并探讨游泳运动对其干预作用及相关机理。方法:5周龄雄性SD大鼠随机分为4组:对照组(C组)、高脂饲料组(HF组)、游泳运动组(CE组)和高脂运动组(HE组),每组8只。C组和CE组喂以普通饲料,HF组和HE组喂以高脂饲料;CE组和HE组进行为期10周无负重游泳运动。10周后测定实验大鼠体重、体长和睾脂重,并计算Lee's指数;腹主动脉取血测定SOD活性和MDA含量;检测精子凋亡率及睾丸组织caspase-3和Cyt c的含量及mRNA水平。结果:1)长期高脂膳食大鼠体重、睾脂、Lee's指数均显著或明显升高;而10周游泳运动后明显下降。2)与C组相比,HF组大鼠精子凋亡率和睾丸组织中caspase-3含量显著升高、血清MDA的含量、睾丸组织Cyt c含量及caspase-3和Cyt c mRNA表达水平明显升高,而血清中SOD活性明显降低;与HF组相比,HE组大鼠精子凋亡率、血清MDA的含量、睾丸组织Cyt c含量明显降低,睾丸组织caspase-3含量显著降低,血清SOD的活性明显增加。结论:长期高脂膳食可诱导大鼠精子凋亡率明显上升,其凋亡机制可能与机体氧自由基增加及caspase-3、Cyt c增多有关;游泳运动可改善高脂诱发的精子凋亡,其改善原因可能是因为激活自由基代谢及caspase-3、Cyt c减少引起的。
Objective: This study focused on the effects of long-term high-fat diet on apoptosis of spermatogenic cells of male SD rats, and explored the effect and mechanism of swimming exercise on apoptosis of spermatogenic ceils in high-fat diet. Methods : Thirty-two five-weeks-old male SD rats were randomly divided into 4 groups ( n = 8) : control group (C group) ,high-fat diet group (HF group) ,exercise group (CE group) and high-fat plus exercise group ( HE group). C and CE groups were fed with normal diet. HF and HE groups were fed with high-fat diet. CE and HE group rats participated in swimming, 10 weeks in total. After 10 weeks the rats'weight, body length and weight of adipose tissue were measured,and Lee's index was calculated. SOD activity and MDA content in blood from abdominal aortic were measured, sperm apoptosis rate, and the mRNA and protein expressions of easpase -3 and Cyt c in testieular tissue were tested. Results: 1 ) After 10 weeks high-fat diet,rats' body weight, adipose tissue weight of testis, and Lee's index increased significantly, but they decreased after 10 weeks swim- ming. 2) Compared with C group, sperm apoptosis rate, the mRNA and protein expressions of caspase - 3, Cyt c in testicular tissue, and MDA content in serum were higher in HF group ,while SOD activity in serum was lower in HF group. Compared with HF group, sperm apoptosis rate, MDA content in serum, and caspase -3 and Cyt c content in testicular tissue were decreased in HE group, while SOD activity in serum increased. Conclusion: Long-term high-fat diet can induce higher sperm apoptosis rate, which may be related to the increases of SOD, caspase - 3, Cyt c. Swimming exercise can control the increase of sperm apoptosis rate induced by high-fat diet, which may because of the decreases of SOD, caspase - 3, Cyt c after swimming.
