摘要
目的:研究c-ski在食管鳞癌组织中的表达以及与临床病理特征之间的关系,探讨食管鳞癌的发病机制并为其早期诊断及治疗寻找新的因子。方法:采用原位杂交法及免疫组化SP法检测22例正常食管组织,24例上皮内瘤变组织及84例食管鳞癌组织中c-ski mRNA及c-ski蛋白的表达情况。结果:c-ski mRNA在食管上皮组织,上皮内瘤变组织和食管鳞癌组织中的阳性表达率分别是13.6%(3/22),62.5%(15/24),92.9%(78/84),3组差异具有高度统计学意义(P<0.01);c-ski蛋白在食管上皮组织,上皮内瘤变组织和食管鳞癌组织中表达阳性率分别是6.67%(6/22),45.8%(11/24),70.2%(59/84),3组差异具有高度统计学意义(P<0.01)。c-ski蛋白在食管鳞癌中的表达与TNM分期有相关性(P<0.05),而与患者的年龄、性别、肿瘤分化程度、淋巴结转移等均关系不大。结论:c-ski可能是一个潜在的癌基因,在食管鳞癌的发生发展中起作用。
Objective: Detect the expression of c-ski in squamous cell carcinoma of esophagus, and investigate the relationship between the expression and clinical pathology of squamous cell carcinoma of esophagus. Explore and discuss the pathogenesis of esophageal squamous cell carcinoma(ESCC) new factors for early diagnosis and treatment. Methods:84 cases of squamous cell carcinoma of esophagus and 24 cases of EIN are regarded as the experimental group and 22 cases of normal esophagus tissues as the control group. Then the expression of c-ski protein in squamous cell carcinoma of esophagus, EIN and normal esophagus tissues by immunohistochemi- cal( IHC,SP method) are detected as well as the expression of c-ski mRNA by hybridization in situ(ISH). Results:The positive ex- pression rates of c-ski mRNA in ESCC ,EIN and normal esophagus tissue are 92.9% (78/84) ,62.5% (15/24) and 13.6% (3/22) ,respectuely. There was statistical significance among the groups(P 〈 0. 01 ). The positive expression rate of c-ski protein in ESCC, EIN and normal esophagus tissue was 70.2% (59/84) ,45.8% ( 11/24 )and 6. 67% (6/22) , resprc tively. There was statistical significance among the groups( P 〈 0.01 ). Expression of c-ski protein in ESCC indicate that there was statistical significance with TNM stages( P 〈 0. 05 ) and no relation with lymph node metastasis, histological grade, age and prognosis ( P 〉 0.05 ). Conclusion : The up-regulated expression of c-ski in ESCC tissue may be related to the development of ESCC. C-ski plays an important role in the metastasis and development of ESCC.
