摘要
目的观察RUNX3在结直肠正常组织、结直肠腺瘤及结直肠腺癌组织中的表达,探讨RUNX3在结直肠正常黏膜→腺瘤→腺癌过程中的表达变化及临床意义。方法采用免疫组化法、荧光原位杂交技术检测RUNX3在30例癌旁正常组织、60例结直肠腺瘤(37例伴中低度异型增生,23例伴重度异型增生)及60例结直肠癌组织中的表达,结合临床资料分析其表达与临床病理参数的关系。结果 RUNX3在结直肠正常组织、腺瘤伴中低度异型增生、腺瘤伴重度异型增生及结直肠癌组织中的阳性率分别为86.67%、72.97%、43.48%、38.33%,差异具有统计学意义(P<0.008 3)。RUNX3 mRNA在结直肠正常组织、腺瘤伴中低度异型增生、腺瘤伴重度异型增生及结直肠癌组织中的阳性率分别为90.00%、78.38%、43.48%和40.00%,差异具有统计学意义(P<0.008 3)。RUNX3表达与结直肠癌TNM分期、浸润深度、淋巴结转移、肝转移、腹腔微转移具有相关性(P<0.05),而与患者性别、年龄、肿瘤大小、部位、分化程度和组织学类型无关(P>0.05))。结论 RUNX3表达与结直肠黏膜癌变的发生、发展密切相关,有望成为结直肠癌诊断及监测的特异性生物学指标,并为结直肠癌的治疗起指导作用。
Purpose To study the expression of RUNX3 in human normal colorectal mucosa, colorectal adenoma and colorectal adenocarcinoma tissues and to explore their correlation with the initiation and progrssion of adeuocarcinoma and its clinical significance. Methods The expression of RUNX3 in 30 cases of normal eolorectal tissues, 60 colorectal adenoma tissues with 37 mild dysplasia ( grade Ⅰ or Ⅱ dysplasia) and 23 severe dysplasia ( grade m dysplasia), and 60 colorectal adenocarcinomas were detected by using immunohistochemical technique and in situ hybridization. The correlation between their expression and the clinicopathologie features was analyzed. Results The positive rates of RUNX3 in 30 normal colorectal tissues were 86. 67%, while in 60 colorectal adenoma tissues with mild dysplasia (37 cases) andsevere dysplasia (23 cases), they were 72. 97% and 43.48%, respectively and in 60 eolorec- tal adenocarcinoma tissues they were 38.33%. The difference was significant (P 〈 0. 008 3 ). The positive rates of RUNX3 mRNA in normal colorectal tissues were 90. 00%, while in 60 colorectal adenoma tissues with mild dysplasia andsevere dysplasia, they were 78.38% and 43.48%, respectively and in 60 colorectal adenocarcinoma tissues they were 40. 00%. The difference was significant (P 〈 0. 008 3 ). Expression levels of RUNX3 were correlated with the TNM staging, liver metastasis, the depth of tumor invasion, lymph node metastasis and peritoneal micrometastasis (P 〈 0. 05 ), but not with the patient's age, gender, site, tumor size, the differentia- tion degree, and histological classification ( P 〉 0. 05 ). Conclusion The high expression levels of RUNX3 have a close correlation with the initiation and progression of colorectal cancer, which has a potential to become a new target for clinical diagnosis and treatment of colorectal cancer.
