摘要
利用RAPD标记技术对山东省克氏原螯虾(Procambarus clarkii)3个地理群体(微山湖、东平湖、济南小清河)的遗传多样性进行了分析。结果表明:在优化的反应条件下,所用12个随机引物共扩增出118个位点,其中48个多态位点。遗传距离显示克氏原螯虾群体间有一定遗传分化,其中济南小清河群体和微山湖群体之间的遗传距离最大(0.3155),微山湖群体和东平湖群体之间的遗传距离最小(0.2027)。济南小清河群体内的遗传相似系数最高(0.9411),微山湖群体内的最低(0.9023)。UPGMA聚类结果显示,微山湖群体和东平湖群体聚为一支,济南小清河群体聚为另一支。
The genetic diversity of threeProcambarus clarkia populations of Shandong Province were analyzed by RAPD.The result showed that under the optimized response condition,totally 118 DNA loci were amplified by 12 RAPD primers,48 of which polymorphic DNA loci.Genetic distance showed that three populations of Procambarus clarkia had some genetic differentiation.The highest inter-population genetic distance among them was Jinan and Weishan populations(0.3155).The lowest was Weishan andDongpingpopulations(0.2027).The hightest intar-population genetic similarity index amongthose 3populations was Jinanpopulation(0.9411),and the lowest was Weishan population(0.9023).Cluster analysis using the UPGMA method showed that the Weishan and Dongping populations clustered first,and then clustered with the Jinan population.
