石油烃降解产甲烷菌系DNA提取方法比较研究 被引量:2

摘要 基因组DNA提取方法是评价微生物种群多样性的关键因素,文章研究了不同提取方法对石油烃降解产甲烷菌群结构的影响。分别采用如下五种方法提取纯化DNA:方法 I:玻璃珠振荡破壁、Tris平衡酚(pH值8.0)、氯仿、异戊醇提取,Promega试剂盒纯化法;方法 II:玻璃珠振荡破壁,水饱和酚(pH值5.2)、氯仿、异戊醇提取,Promega试剂盒纯化法;方法 III:进口试剂盒1+核酸自动提取仪法,方法 IV:玻璃珠振荡破壁,核酸自动提取仪提取法;方法 V:国产试剂盒提取法。通过末端限制性片段长度多态性(T-RFLP)分析细菌和古菌群落结构组成。采用不同方法获得的细菌和古菌的主要菌群结构类似,但是相对丰度存在差异。 The method of genomic DNA extraction is a key factor to evaluate microbe community diversity. Five methods of extracting and purifying DNA were experimented in this study: method I : beads-beating, Tris saturated phenol( pH 8.0), chloroform, isoamylol extraction, Promega DNA purify kit; method Ⅱ : beads-beating, water saturated phenol ( pH 5.2 ), chloroform, isoamylol extraction, Promega DNA purify kit; method Ⅲ : nucleic acid automatic extraction method; method IV : beads-beating, nucleic acid automatic extraction method; method V : domestic kit method. The T-RFLP was adopted to analysis the composition of bacteria and archaea community. The result showed that community structures obtained by different method were similar, but the relative abundance were different.
