电针手足阳明经穴对丙泊酚麻醉大鼠学习记忆力的影响

Electroacupuncture at Hand and Foot-Yangming Meridian Improves Learning and Memory Abilities of Rats Anesthetized by Propofol

摘要 目的:探讨电针手足阳明经穴对丙泊酚麻醉大鼠学习记忆力的影响及其作用机制。方法:将24只SD雄性大鼠随机分成3组,即空白组(生理盐水+电针非穴位)、药物组(丙泊酚+电针非穴位)、治疗组(丙泊酚+电针阳明经穴);观察各组大鼠Morris水迷宫实验中学习记忆力的变化,同时检测大鼠海马区胆碱酯酶(AChE)和胆碱乙酰化酶(ChAT)活性变化。结果:药物组大鼠第一象限路程和穿越平台区域次数显著少于空白组(P<0.05),治疗组在这两方面均多于药物组(P<0.05),治疗组与空白组之间无显著性差异(P>0.05)。与空白组比较,药物组大鼠海马AChE活性上升(P<0.05)、ChAT活性下降(P<0.05);与药物组比较,治疗组AChE活性下降(P<0.05)、ChAT活性上升(P<0.05);治疗组与空白组间两者无显著差异(P>0.05)。结论:电针手足阳明经穴可改善丙泊酚麻醉所致的大鼠学习记忆力减退,其机制可能与调节海马区胆碱能系统有关。 Objective: To investigate the effect of electroacupuncture (EA) at hand and foot-Yangming meridian on learning and memory abilities of rats anesthetized by propofol. Methods: Twenty-four SD male rats were randomly divided into 3 groups: control group (saline + EA at non-acupoint) , model group (propofol + EA at non-acupoint) , and acupuncture treatment group (propofol + EA at hand and foot-Yangming Meridian). Morris water maze test (MWMT) was conducted to investigate the learning and memory abilities of rats. Activities of aeetyeholinesterase (AchE) and choline acetyltransferase (CHAT) in hippocampus were measured. Results: In comparison to control group, swimming distance in target quadrant as well as passing times over platform was significantly reduced in model group, which were both improved by EA at hand and foot-Yangming meridian in acupuncture treatment group, without significant difference between treatment group and blank group ( P 〉 0.05 ). Compared with blank group, the hippocampal AchE activity was significantly increased ( P 〈 0.05 ), while ChAT activity was reduced in model group (P 〈 0.05). Compared with medicine group, the activity of AchE was decreased in treatment group ( P 〈 0.05 ), and activity of ChAT was increased ( P 〈 0.05 ) ; without significant difference between the two groups ( P 〉 0.05 ). Conclusion : EA at hand and foot-Yangming meridian can improve learning and memory abilities in rats anesthetized by propofol, its mechinsm may be association with the regulation cholinergic system in hippocampus.
