摘要
目的:探讨人参皂苷Rb1抑制长春新碱诱导的小鼠骨髓基质细胞凋亡的作用。方法:研究分为正常对照组、长春新碱组、长春新碱凋亡诱导组,不同浓度人参皂苷Rb1(1.0、5.0、10.0、15.0μg/ml)抗长春新碱(5.0μg/ml)对骨髓基质细胞的生长抑制和凋亡组;采用MTT法测定骨髓基质细胞增殖,采用Annexin V/PI双染流式细胞术及Hoechst 33342荧光染色法测定细胞凋亡。结果:人参皂苷Rb1在5.0~15.0μg/ml浓度范围内显著地降低了长春新碱对骨髓基质细胞的生长抑制;人参皂苷Rb1在5.0~15.0μg/ml浓度范围内显著地阻断长春新碱对骨髓基质细胞的生长抑制。人参皂苷Rb1抗长春新碱诱导的骨髓基质细胞凋亡具有浓度依赖性。结论:人参皂苷Rb1能够抑制长春新碱诱导的骨髓基质细胞凋亡。
Objective: To explore the effect of Ginsenoside Rb1( GS Rb1) against the apoptosis of mice bone marrow stromal cells( BMSCs) induced by vincristine( VCR). Methods: The study was divided into normal control,VCR group,VCR apoptosis group and GS Rb1( 1. 0,5.0,10. 0,15. 0 μg / ml) experiment groups. The BMSCs proliferation was determined using MTT assay. The BMSCs apoptosis was determined using fluorescence-activated cell sorter( FACS) with Annexin V /PI and fluorescent staining method with Hoechst 33342. Results: The VCR( 5.0 μg/ml)-inhibited BMSCs proliferation was promoted by GS Rb1 from 5.0 to 15.0 μg/ml( P <0.05). The VCR( 5.0 μg/ml)-induced BMSCs apoptosis was inhibited by GS Rb1 from 5. 0 to 15. 0 μg /ml( P < 0. 05) and threr was relation of dose-dependent in the effect of GS Rb1 against VCR-induced BMSCs apoptosis. Conclusion: The Ginsenoside Rb1 is able to inhibit VCR-induced BMSCs apoptosis.
