橙皮苷对功能性消化不良大鼠胃肠运动及Ghrelin表达的影响 被引量:40

Effects of hesperidin on functional dyspepsia gastrointestinal motility of rats and the expression of Ghrelin

摘要 目的:观察橙皮苷对功能性消化不良(Functional Dyspesia,FD)大鼠胃肠运动及Ghrelin表达的影响,并进一步探讨其可能的作用机制。方法:用强迫游泳法制备功能性消化不良大鼠模型,观察橙皮苷对功能性消化不良大鼠胃肠运动功能的影响;Elisa法检测血清中Ghrelin表达的改变;免疫组化染色观察大鼠大脑、胃、肠组织中Ghrelin的表达;Western blot法检测大鼠海马Ghrelin蛋白的表达。结果:与正常对照组相比,模型组大鼠胃排空率及小肠推进率明显降低,脑、胃、小肠组织中Ghrelin的表达明显降低。与模型组相比,莫沙必利10 mg/kg、橙皮苷10、40 mg/kg能够改善FD模型大鼠的胃肠运动功能及脑、胃、小肠组织中Ghrelin的表达。结论:橙皮苷能够明显改善功能性消化不良大鼠的胃肠功能,这可能与其增加了血清胃、肠、脑组织中Ghrelin的含量有关。 Objective: To investigate the effects of hesperidin on functional dyspepsia( FD) gastrointestinal motility of rats and the expression of Ghrelin and the possibleand mechanisms. Methods: Preparation of FD model of rats by forced swimming method,observe the effect of hesperidin on gastrointestinal motor function of FD rats. Change of Ghrelin in serum was assayed by Elisa. Immunohistochemical staining observe the expression of Ghrelin in brain,stomach,intestinal tissue in rats. To detect the expression of Ghrelin protein in hippocampus of rats with Western blot methed. Results: Compared with the normal control group,model group rats of the rate of gastric emptying and small intestinal propulsion rate were obviously decreased,the expression of Ghrelin in brain,stomach,small intestine tissues were decreased. Compared with the model group,mosapride 10 mg /kg,po and hesperidin 10,40 mg /kg,po could improve the FD model rat of gastrointestinal function and the expression of Ghrelin in brain,stomach,intestinal tissues. Conclusion: Hesperidin can significantly improve the functional dyspepsia rats gastrointestinal function,this may be related with the increase of Ghrelin in serum,stomach,intestine,brain tissue content.
