红景天苷改善胰岛素抵抗HepG2细胞糖代谢及分子机制初探 被引量:14

Effect and molecular mechanism of salidroside improving glucose metabolism in HepG2 induced by insulin resisitance

摘要 目的:研究红景天苷对胰岛素抵抗HepG2细胞糖代谢的影响以其可能的分子机制。方法:采用1×10-6mol/L高浓度胰岛素诱导HepG2细胞产生胰岛素抵抗模型,葡萄糖氧化酶法和蒽酮法分别检测红景天苷对模型细胞葡萄糖利用、糖原合成的影响,RT-PCR法检测胰岛素抵抗相关基因IRS-2、GLUT-2、PPAR-γmRNA的表达。结果:与模型组相比,浓度为1×10-6mol/L的红景天苷可促进胰岛素抵抗HepG2细胞葡萄糖的利用和糖原合成,IRS-2 mRNA表达明显升高,GLUT-2 mRNA显著降低,PPAR-γmRNA表达没有显著改变。结论:红景天苷可以改善胰岛素抵抗细胞糖代谢,其机制可能与升高IRS-2,降低GLUT-2 mRNA有关。 Objective :To explore the effect and molecular mechanism of Salidroside improving glucose metabolism in HepG2 induced by insulin resisitance. Method :The insulin resistant models of HepG2 were induced by the 1 × 10^-6mol/L high concentnifion of insulin for 36 h. We evaluated glucose utilization and glycogen synthesis of the model cells by glucose oxidase enzyme kit and anthrene assay respectively, RT-PCR assay was empolyed to analyze mRNA expression of IRS-2, GLUT-2 and PPAR-γ. Results:Compared with model group, the 1× 10^-6mol/L concentration of Salidroside can promote insulin resistance cell glucose Utilization and glycogen synthesis. The mRNA expression of IRS-2 was signifaneant higer than model group(P 〈 0.05), then GLUT-2 mRNA decreased(P 〈0. 05), PPAR-γ mBNA expression did not change significantly. Conclusion: Salidroside improved glucose metabolism in sulin resistance HepG2, which was associated with elevated IBS-2, reduced GLUT-2 expression.
