摘要
目的探讨黄芪多糖(astragalus polydsaccharide,APS)对视网膜母细胞瘤RB44细胞侵袭能力的影响及机制。方法以200 mg·L-1APS诱导48 h的RB44细胞为实验组(预实验结果),以不加药的RB44细胞为对照组。通过Transwell迁移和侵袭实验观察APS对RB44细胞迁移和侵袭能力的影响,通过Western blotting方法检测APS处理前后RB44细胞中侵袭相关基因蛋白表达的变化。结果 Transwell迁移和侵袭实验结果显示:与对照组每高倍视野(54.667±4.041)个和(26.000±2.646)个细胞相比,APS组RB44细胞穿过Transwell小室基膜的细胞数量显著减少,分别为每高倍视野(26.667±4.041)个和(8.667±2.082)个细胞(均为P<0.01)。Western blotting结果显示:与对照组(0.128±0.019)相比,APS组RB44细胞中E-cadherin的表达(0.117±0.019)差异无统计学意义(P=0.486),而MMP-9和MMP-2的表达(0.134±0.008、0.085±0.006)与对照组(0.269±0.187、0.223±0.013)相比均显著降低,差异均有显著统计学意义(均为P<0.01)。结论 APS可显著抑制视网膜母细胞瘤RB44细胞的侵袭能力,其机制可能与抑制MMP-9和MMP-2的表达有关。
Objective To explore the effects of astragalus polysaccharide (APS) on the invasive potential of retinoblastoma cells and it mechanisms. Methods Taken RB44 cells untreated with APS as the control group, RB44 cells treated with 200 mg · L ^- 1 APS for 48 hours as the APS group. The influence of APS on the migration and invasion potential of RB44 cells was determined via transwell migration test and transwell invasion test. The changes of invasion related genes before and after treated with APS were detected by Western blotting assay. Results The results of transwell migration test and transwell invasion test revealed that, compared with control group ( 54. 557 ±4. 041 ) cell/HPF and ( 25. 000 ±2. 545 ) cell/HPF number of RB44 cells through basement membrane of chamber in APS group ( 25.557 ±4. 041 ) celVHPF and ( 8. 557 ±2. 082 ) cell/HPF were significantly decreased ( both P 〈 0.01 ). The results from Western blotting demonstrated that, compared with expression of E-cadherin in control group (0. 128 ±0. 019) ,the expression of E-cadherin in APS group (0. 117 ±0.019) was no significant difference ( P 〉 0. 05 ), while the expressions of MMP-9 and MMP-2 in APS group (0.134 ±0. 008,0. 085 ±0. 005 ) were significantly increased compared with control group(0. 259 ±0. 187,0. 223 ±0. 013) (both P 〈0.01). Conclusion APS could remarkably inhibit the invasive ability of retinoblastoma RB44 cells, which the mechanisms may be related to the inhibition of APS on the expressions of MMP-9 and MMP-2.
