7型腺病毒中和多肽的筛选及其活性研究

摘要 目的筛选7型腺病毒的结合多肽,并研究其对7型腺病毒的结合和中和活性。方法利用噬菌体肽库,筛选7型腺病毒的结合多肽,非竞争ELISA法测亲和常数测定与7型腺病毒的亲和常数,微量中和试验测定对7型腺病毒的中和活性。结果筛选到了GTS09多肽,其与噬菌体的复合物(GTS09-phage)能特异的结合7型腺病毒,并且能够高效的中和7型腺病毒,GTS09与BSA的复合物也有中和作用,但GTS09本身却没有中和活性。结论大分子蛋白可能会改变GTS09的空间构象,从而使其具有中和7型腺病毒的能力。 Hu man adenovirus serotype 3 (Ad3) and serotype 7 (Ad7) are important pathogens causing respiratory tract diseases, such as acute respiratory disease in pediatric and adult patients, and no specific therapeutic drugs can be used so far. In this study, a peptide, named GTS09, was found by panning a random peptide libraries against Ad7. And the complex (GTS09-phage and GTS09-BSA) could bind and neutralize Ad7 efficiently, while GTS09 itself could not. These results show that macromolecular protein may alter the conformation of GTS09 so that it has the ability to neutralize Ad7.
