摘要 病理性的血管重建是一类由多种因子和多种细胞参与的复杂过程,在动脉粥样硬化、高血压等心血管疾病中普遍存在。组蛋白去乙酰化酶(HDACs)作为一类重要的染色体结构调节酶类,几乎参与所有的细胞生化过程。传统的HDACs研究主要集中在其抗肿瘤的作用上,但近来有大量报道表明,HDACs在血管重建中也起到重要的作用。本综述总结了HDACs与新生血管形成的关系,包括其对内皮细胞的增殖、迁移、生存和分化的影响。此外,HDACs的抑制剂具有较明显的抑制新生血管形成的作用,是一种全新的肿瘤治疗和抗血管增生途径。因此,探讨HDACs参与血管重建的机制将为动脉粥样硬化、高血压等心血管疾病的治疗提供新的方向。 Pathological vascular remodelling is a complex process involved in multiple factors and cells. And it is prevalent in cardiovascular disease such as atherosclerosis, hypertension, and etc. Histone deacetylases (HDACs)play critical roles in almost all the biological processes via modulating chromatin structure. Recently, mounting evi dence has indicated HDACs are important for vascular remodelling, although original reports suggested HDACs were involved in cancer and neurological diseases. This review summarized the effects of HDACs on angiogenesis, a process including endothelial cell proliferation, migration, survival and differentiation. Alternatively, we also re- vealed the potential therapeutic implications in anti-tumour and anti-angiogenesis due to the effects of HDAC antago- nists on inhibiting neovascularization. Accordingly, the investigation on the mechanisms of HDACs involved in vas- cular remodelling will provide promising therapeutic targets for the treatment of arteriosclerosis, hypertension, and other cardiovascular diseases.
