摘要
目的依据Mesotherapy的治疗理念,观察真皮内注射二甲氨基乙醇(DMAE)和复方氨基酸溶液(AA)对D-半乳糖诱导的化学性衰老大鼠皮肤MMP-1和TIMP-1mRNA表达的影响。方法80只wistar大鼠随机分人正常对照组、衰老对照组、衰老治疗组。衰老治疗组从注射D.半乳糖后第18天开始,在大鼠两侧臀部皮肤分别皮内注射0.2%DMAE+AA、0.1%DMAE+AA、0.2%DMAE、0.1%DMAE、AA、生理盐水(NS),每周1次,连续4周。观察各组皮肤组织学、真皮厚度、羟脯氨酸(Hyp)、MMP-1、TIMP-1mRNA和成纤维细胞核增殖抗体(PCNA)表达水平的变化。结果真皮内注射0.1%DMAE+AA、0.2%DMAE+AA,可使衰老大鼠真皮厚度、Hyp含量和MMP-1mRNA表达水平显著增加,而对TIMP-1mRNA和PCNA的表达没有影响。结论在衰老大鼠真皮内注射0.2%DMAE+AA、0.1%DMAE+AA,可促进老化、变性胶原蛋白分解,重构衰老皮肤组织结构,使皮肤中胶原蛋白含量显著增多,真皮厚度增加.具有一定延缓皮肤衰老的作用。
Objective To study the effect of dimethylaminoethano and compound amino acid on MMP-1 and TIMP-1 mRNA expression of rat aging skin induced by D-galactose. Methods Eighty wistar rats were ran- domly divided into the normal control group, the aging control group and the experimental groups. At 18 days af- ter D-gal induction, the experimental groups were treated with intradermal microinjeetion of 0.2% DMAE + AA, 0. 1%DMAE + AA,0. 2% DMAE,0.1% DMAE,AA and normal saline (NS) on bilateral buttocks respectively, once a week for 4 weeks. At 42 days after treatment, the histological changes, the dermal thickness, hydroxypro- line (Hyp) and mRNA expression of MMP-1, TIMP-1 and PCNA in every group were detected. Results Meso- therapy with intradermal co-injection of 0.1% DMAE + AA, or 0.2% DMAE + AA significantly increased the der- mal thickness, Hydroxyproline content and MMP-1 mRNA expression levels, but did not affect the expression of TIMP-1 mRNA and PCNA in aging rats. Conclusion Local co-injection of DMAE and compound AA directly into target tissue under mesotherapy has marked anti-aging effects on D-galactose induced skin aging model of rat by degradation of aging collagen fiber and reconstruction of skin structure.
