摘要
目的:研究白薇经皮透过液对B16黑色素瘤细胞增殖的抑制效果,初步探讨白薇作为皮肤美白剂的可行性。方法:以每孔3×103个B16细胞接种于96孔板,分别加入质量浓度为20,60,100,140,180,200 mg·L-1的白薇经皮透过液,作用24,48,72 h,采用MTT法测定白薇经皮透过液对B16细胞增殖的影响;L-Dopa氧化法测定透过液对B16细胞内酪氨酸酶(TYR)活性的影响;NaOH裂解法测定透过液对黑色素含量的影响;4',6二脒基-2-苯基吲哚(DAPI)染色法及原位末端转移酶标记染色(TUNEL)法检测细胞凋亡。结果:与空白组相比,不同浓度不同提取方法的白薇经皮透过液均对B16细胞的增殖、酪氨酸酶的活性及黑色素的含量具有抑制作用(P<0.05),作用效果:95%醇提>50%醇提>水提,最佳作用质量浓度为200mg·L-1,最佳作用时间为48 h。结论:白薇具有一定的美白作用,可用于美白产品的开发。
Objective: Tostudy the effect on the proliferation of the B16 melanoma cell, preliminarily discuss the feasibility of Cynanchi Atrati Radix et Rhizoma as a skin whitening agent. Method: B16 cells 3 × 10-3 were seeded in 96-well plates which were added at a concentration of 20, 60, 100, 140, 180, 200 mg .L-lof Cynanchi Atrati Radix et Rhizoma percutaneous fluid. The proliferation, tyrosinase activity and melanin biosynthesis of the B16 melanoma cell were measured by MTT cytotoxicity assay, utilization of L-dopa and NaOH-assay respectively; 4', 6-diamidino-2-phenylindde (DAPI) staining and TUNEL method to detect apoptosis. Result: Compared with the control group, different concentrations of different extraction methods of Cynanchi Atrati Radix et Rhizoma percutaneous liquid through the proliferation of B16, tyrosine enzyme activity and melanin content had inhibitory effect (P 〈0. 05) , the sequence was: 95% alcohol extract 〉 50% alcohol extract 〉 water extract, the best concentration was 200 mg·L-1, the best reaction time was 48 h. Conclusion: Cynanchi Atrati Radix et Rhizoma has whitening effect, and can be used for whitening products.
