摘要
目的:考察渴络欣胶囊(KLX)对糖尿病视网膜病变的防治作用。方法:采用腹腔注射链尿佐菌素(STZ)建立糖尿病大鼠模型,4周后筛选血糖值稳定的模型大鼠80只(血糖值≥15.8 mmol·L-1)采取预防模式给药,另取首次注射3个月后的模型大鼠80只[血糖≥15.8 mmol·L-1,视网膜中央动脉(CRA)血流量明显降低]采取治疗模式给药。预防和治疗模式均随机分为模型组、多贝斯组(0.5 g·kg-1)及KLX高、中、低剂量组(2.0,1.0,0.5 g·kg-1);另各设正常对照组10只,ig 1次/d,预防模式连续给药21周,治疗模式连续给药12周,末次给药后检测大鼠视网膜中央动脉(CRA)血流动力学、全血黏度、血沉、血浆中血管内皮生长因子(VEGF)、组织病理学和视网膜血管内周细胞数和内皮细胞数变化。结果:采用预防和治疗模式给药,KLX各剂量组均能显著增加模型大鼠视网膜CRA血流流速,降低大鼠全血黏度、血浆黏度;减少视网膜血管内周细胞的丢失和抑制内皮细胞的增生,减轻视网膜水肿程度,改善视网膜血管壁变形或血管壁细胞排列不规则。各剂量组均能不同程度的降低血浆中VEGF。结论:KLX对大鼠糖尿病性视网膜病变具有显著的防治作用。
Objective: To investigate the preventive and therapeutical effects of Keluoxin caspsule (KLX) on diabetic rats with diabetic retinopathy. Method: Heathy Sprague-Dawley (SD) rats were induced by intraperitoneal injection of streptozotocin (STZ, 50 mg ·kg-1). Four weeks later, eighty rats with stable blood glucose ( ≥ 15.8 mmol ·L-1) were selected as preventive mode. Other eighty diabetic retinopathy (DR) rats with stable blood glucose (I〉15.8 mmol ·L-1) and lower central retinal artery (CRA) were choose as therapeutical mode after three month. The 180 rats of both modes were divided randomly into control group, calcium dobesilate (CAD) group (0.5 g·kg-1) and KLX group (2.0, 1.0, 0.5 g·kg-1), ten healthy rats were used as normal group. The model rats was gavaged once a day and the effect of KLX was studied via detecting CRA, blood viscosity, erythrocyte sedimentation rate, vascular endothelial growth factor (VEGF) concentration in plasma, endothelia/pericyte (E/P) value and histopathology after the final administration. Result: Both in preventive and therapeutic KLX groups, the whole blood and plasma specific viscosity, E/P value decreased significantly, and the blood supply of CRA improved obviously. Even more important, KLX could extenuate retinal edema, improve deformation of retinal wall cell in STZ induced diabetic retinopathy rats. Meanwhile, therapeutical group (2.0 g·kg-1) can decrease plasma VEGF marked preventive and therapeutical effects on occurrence and the the pathological histology analysis showed that blood vessels and irregular arrangement of vessel preventive groups (0.5, 1.0 g ·kg-1) and level to some extent. Conclusion: KLX exert development of diabetic retinopathy.
