摘要
目的:研究蒙药库页悬钩子中总皂苷和齐墩果酸的含量测定方法及最佳采收期。方法:选用齐墩果酸为对照品,采用分光光度法,5%香草醛-冰醋酸和高氯酸显色,在波长547nm处测定吸光度,计算不同采收期库页悬钩子中总皂苷含量;采用高效液相色谱法对齐墩果酸含量进行测定,Phenomsil C_18色谱柱(4.6mm×250mm,5μm),柱温为室温(25℃),流动相甲醇-0.3%磷酸水溶液(80:20),流速1mL·min-1,检测波长220nm。结果:不同采收期库页悬钩子中总皂苷和齐墩果酸含量发生明显变化。总皂苷在6-9月含量逐渐升高,9月含量最高,10月逐渐下降,齐墩果酸含量从8月开始逐渐升高,9月达最大值,10月下降。结论:该实验方法简便、快速、重复性好,可用于库页悬钩子总皂苷和齐墩果酸的含量测定,库页悬钩子宜于9月采集。
Objective: To establish the content determination method for the total saponinsandoleanolic acid of Rubus sachalinensis and the best collection stage. Method: Using the UV-visible spectrophotometry determination the content of total saponins at wavelength of 547 nm and using 5% vanillin-acetie acid and perchloric acid coloration. An HPLC method for oleanolic acid of R. sachalinensis was established to determineoleanolic acid. Result: There were differences in the content of the total saponinsand oleanolie acid of R. sachalinensis at different collecting time; the content of total saponins increased from June to September and reached the highest level in September. However, it gradually decreased in October. Conclusion: The method was simple, rapid, repeatableand could be used for the total saponins and oleanolie acid determination of R. sachalinensis, the best collection stage was September.
