摘要
目的研究黄芩甙对脑缺血再灌注后损伤中凋亡因子表达的影响。方法选用Wistar大鼠进行脑缺血再灌注造模,利用免疫组化等方法,检测经黄芩甙治疗后鼠脑半暗带区JNK、ERK1/2和p38的表达含量变化。结果黄芩甙可以降低神经功能评分,与盐水治疗组对比,于6 h时的2.17±0.75降低到1.50±0.55,同时可分别下调缺血再灌注后JNK、ERK1/2和p38的表达,ERK1/2表达于2 h、6 h和12 h时比盐水治疗组下降,JNK表达于各个时间点均下降,而p38的表达除了在6 h时其他各个时间点均下调,均有统计学意义(P<0.05)。结论黄芩甙通过抑制JNK、ERK1/2和p38等凋亡基因能够减少神经元凋亡和降低神经功能缺损。
Objective To study the effects of baicalin on expression of apoptosis factors after brain ischemia reperfusion. Methods Wistar rats were used to establish cerebral ischemia reperfusion model, and expression changes of JNK, ERK1/2 and p38 in rat brain penumbra after treated with baicalin was test by immunohistochemical method. Results Baicalin could decrease the neurological function score (NFS), compared with model group, the NFS in 6 h (2. 17 ±0. 75) reduced to ( 1.50 ±0.55) , and expression of JNK, ERK1/2 and p38 were down regulated after ischemia reperfusion, and the expression of ERK1/2 in 2 h, 6 h and 12 h were increased than saline treated group, and the expression of JNK in different time point were decreased, while the expression of p38 was decreased except for 6 h (P 〈 0. 05 ). Conclusion Baicalin can reduce the apoptosis of neuron and defect neural function injury by inhibiting JNK, ERK1/2 and p38 apoptosis gene.
