电针预处理对大鼠脑缺血再灌注损伤后颈静脉血糖及脑水肿的影响 被引量:5

Effects of electroacupuncture preconditioning on jugular venous blood glucose and brain edema induced by cerebral ischemia reperfusion in rats

摘要 目的观察电针预处理对大鼠脑缺血再灌注损伤后颈静脉血糖和脑组织含水量的影响。方法健康雄性Wister大鼠90只,随机分为假手术组(SH组,n=30只),脑缺血再灌注组(IR组,n=30只),电针预处理加脑缺血再灌注组(EA组,n=30只)。采用阻断双侧颈总动脉合并全身低血压方法建立大鼠全脑缺血模型,缺血15 min实施脑血流再灌注。电针预处理组在脑缺血开始前给予电针刺激穴位预处理30 min,穴位选择"百会"(Du20)与"水沟"(Du26)穴。在缺血再灌注后2、6、24 h,从颈静脉抽取血样测定血糖含量,然后断头取血,采用干-湿重法测定大鼠脑组织含水量。结果 IR组和SH组以及EA组的脑含水量在术后2 h无明显差别。与SH组比较,IR组和EA组脑含水量在再灌注后6 h明显升高(P<0.01)。EA组在再灌注后6 h和24 h脑含水量显著低于IR组(P<0.01)。和SH组比较,EA组和IR组在再灌注后2 h血糖明显升高,6 h达到高峰至24 h后有所回落(P<0.01)。EA组在再灌注2 h,6 h和24 h血糖值显著低于IR组(P<0.01)。结论电针预处理能够显著降低脑缺血灌注大鼠血糖上升水平,可以减轻脑缺血再灌注后脑水肿,从而减轻脑缺血再灌注损伤。 Objective To investigate the effects of electroacupuncture preconditioning on jugular venous blood glucose and brain edema induced by cerebral ischemia reperfusion in rats. Methods Ninety male Wistar rats were randomly assigned to sham operation group( SH,n = 30), cerebral ischemia reperfusion group( IR,n = 30),and electroacupuncture preconditioning group( EA, n = 30). The rat model of transient general brain ischemia w as induced by bilateral common carotid arteries occlusion combined w ith low blood pressure. Cerebral blood reperfusion began after 15 min of cerebral ischemia. Electroacupuncture w as applied to acupoints Baihui( Du20) and Shuigou( Du26) for 30 min immediately before cerebral blood reperfusion in EA group. Jugular vein w as cannulated to collect blood samples for blood glucose analysis at 2、6、24 h after ischemia,respectively. The animals w ere sacrificed at 2、6、24 h after reperfusion,respectively. Brains w ere immediately removed for determination of brain w ater content by the dry-w et w eight method. Results There w as no significant difference in brain w ater contents at 2 h after reperfusion among all groups. Compared w ith SH group, brain w ater content w ere increased significantly at 6 h,peaked at 24 h after reperfusion in IR group and in EA group( P &lt; 0. 01). Brain w ater contents w ere significantly higher in IR group than those in EA group at 6 h,24 h after reperfusion( P &lt; 0. 01). Compared w ith SH group,the levels of the jugular venous blood glucose were increased remarkably at 2 h,peaked at 6 h and continued until 24 h after reperfusion in IR group and in EA group( P &lt; 0. 01),while the levels in EA group were significantly lower than those in IR group( P &lt; 0. 01). Conclusion Electroacupuncture preconditioning can inhibit the increasing of blood glucose and brain water contents,so as to reduce the damage and protect neurons in rats undergoing cerebral ischemia reperfusion.
